以应激蛋白为生物标志物研究低浓度2-硝基-4'-羟基二苯胺对鲫鱼肝脏和脑组织的动态暴露的影响

摘要:
采用实验室模拟方法,以鲫鱼(Carassiusauratus)作为实验动物,运用SDS-PAGE和WesternBlotting方法,研究了在不同暴露时期,1mg/L2-硝基-4'-羟基二苯胺对鲫鱼肝脏和脑组织应激蛋白HSF70诱导表达的情况.结果表明:应激蛋白HSP70对2-硝基-4'-羟基二苯胺非常敏感.在鲫鱼暴露2d后,脑组织中HSP70与对照组相比,有显著的表达(P<0.05);在鱼体暴露7d后,肝脏组织中HSPT0与对照组有显著的诱导表达(P<0.05);随着暴露天数的增加,肝脏组织和脑组织HSP70表达也显著增加,且存在着较好的时间-效应关系.可考虑将应激蛋白HSP70作为2-硝基-4'-羟基二苯胺污染在水生生态系统中的一种敏感的生物标志物.

Dynamic Effects of Low-level 2-nitro4'-hydroxydiphenylamine on the Induction of HSP70 in the Fish Livers and Brains

Abstract:
Carassius awratus was chosen as the experimental materials to study the dynamic effects of 1 mg/L 2-nitro4'-hydroxydi-phenylamine on the induction of HSP70 in the fish livers and brains.Using the SDS-PAGE and Western Blotting techniques, the expression level of HSP70 in the fish livers and brains was evaluated.As shown from the results, 2-nitro-4'-hydroxydiphenylamine treatment resulted in a time-dependent elevation in HSP70 expression at 2, 4, 7, 10 and 16 days exposure.Comparing with the control group,the HSP70 in the fish brains and livers began to express significantly after being exposed for4 days and 7 days respectively.The expression level of HSP70 was enhanced with the exposure days increasing and showed a good time-effect relationship.Results from this study suggest that HSP70 as an environ menial biomarker can be a sensitive and rapid indicator in the aquatic ecosystem.
